You can assign 2 samples to 1 pad via the Program menu. Select the pad under there then to go one of the sub menus (i think its f4) and there should be an option that says "Pad triggers 2 samples" and you select another sample that is loaded on another pad. Check the manual for the specifics.
Also, like beets said for the filter, go shift+after, assign it to a note and move the slider ... but the way I read it, you want real time filtering AFTER the note starts playing ... so you can, say, change the filter on a vocal sample during its playback. Unless its a hidden feature of the MPC2000XL that I haven't found, or I'm just stupid, you can only adjust the filter for 1 playback of the sample you're filtering. You can't change the filter in mid-playback (unless you buy the analog filter thing).
