i use:
mpc2000 og (notXL)
behringer eurorack MX1604 12 channel mixer
fatar midi controller keyboard
electrix moFx multifx unit as a midi knob controller
software:
reason
nuendo
sound forge
recycle
granulab
a mixer is very helpful, especially if you use any other sources like guitars, bass, vocals, etc.
soundforge is great for sample manipulation, slicing, etc
recycle is good for slicing
you can make everything with just an mpc, but having more tools doesnt hurt.

i track audio into nuendo. by audio i mean guitar and vocals and bass and turntables, whatever. i just use the mpc for beats and when it comes time either record that into nuendo, or take the midi file and load it into reason (if I feel like using some different drum sounds and changing things up alot).
