Exchange tips and tricks for the Akai MPC4000

I've played around with the different filters on the MPC 4000 (2-pole, 4-pole, 6-pole, etc.) to filter out the lows of a sample in order to make a bassline. My problem is, it sounds too muffled. I mean, I can layer it over the original sample and it gives it nice depth, but it still doesn't give me that nice sounding crisp bassline. Can somebody help me in achieving this? Do I need to re-filter the sample? Also, if a sample was recorded and it has major hiss, what's the best way to remove that hiss and clear up the sample? Thanks to anyybody that can help....
