Possibly Dumb Hard Drive Question ... (I searched Though)

I got the hard drive installed loaded samples, songs, etc from computer using USB (all thanks to this site pretty much)

I have a question I wanted to load from hard disk because I have a bunch of programs I am making etc

Can I make programs and have samples open when I turn on my MPC straight from my hard drive? If so how?
