Is it possible to:

Play through and sample a synth

that is currently playing in the sequence by MIDI

so you can save that phrase locally to the MP as a sample

?

yes, this is possible: route the audio output of your synth to the mpc in, be sure to be on another track of your sequence and choose or make a program with at least one empty pad.
now move your cursor on the main screen to the "drum" field and press the window key - this will enter direct record mode.
now for conveniance start your recording with rec+play to get a precount - while the precount is runing hit f6 to start the direct recording.
if your sequence has played hit f6 again to stop the recording.
on this screen you can rename your sample and just hit the pad you want to assign it to.
if you choose a pad another option will show up "record event" which defaults to "yes" and you should keep it that way so your sample will be automagically placed right in your sequence and you don“t have to place the sample manually.
you can of course do all that if you want to.

it works, but i've always found it easier to record into my daw then export via usb to the mpc. only takes two or three minutes and you have much greater control over the sound.

or just use JJOS and switch to track 57 or higher and record your sample in sync with the rest of your beat as an audio track...
