mono is useful if you want to press a pad repeatedly and have it cut itself off.
mute groups let you assign several pads to a group (group 1 for example), every time you hit pad in that group it will cut off another pad in that group. you can have lots of mute groups.....i especially use this when i chop up loops - say i have a 2 bar piano loop, i might slice it into 8 parts, then i assign each sample to 8 pads, i add all those pads to a mute group, now i can play those pads and cut each other off and only effect those 8 pads and not the rest of my sounds.
ive also used mute groups for intros. say i have some goofy quote that is like 10 seconds long that i want for my intro. i can assign that intro to group 4, and the say on the first beat of my sequence my drum kit hits, i can assign that to group 4. now i can play the intro, then start playing my sequence and cut off the intro whenever i like......
