Best bet is to use a DI box (do a google search on these) or use a line out from your bass amp, the signal direct from the bass itself will be far too weak without a preamp or amp

Separating the sounds is a tricky business involving use of eq and other effects. There have been loads of posts before about this, I suggest you search this forum. Nothing beats finding "clean" samples without any other sounds in the mix to sample. Altering the sound can be achieved by using filters, eq and effects - read the manual on this and do tons of research, there are loads of websites that help, not least this one (again, use the search function and keep reading other posts) and tweakheadz to name but two. I heard the mpc500 tutorial is worth the money too. And finally, I will repeat what every noob has drummed into them, read the effing manual! :D
