The 5k already has an Autoload folder. If you are just trying to skip the autoload then press MODE+PAD3 to enter DISK mode, and then press UTIL (F3), you can disable Autoload there. This does not delete any samples or sequences, it only prevents them from loading at startup.
If you are looking to delete the samples/sequences permanently (so they are completely gone from the machine) then you just connect your PC to your MPC with a USB cable and then press MODE+PAD3 to enter DISK mode, and then press USB (F4), and then press MOUNT (F6). Once the device shows as connected, just access the 5k like any other drive on your computer...then delete the files normally (on your PC) and dismount the 5k (via the "Safely remove hardware" feature, if you're on windows). Once you've deleted the files, they are gone and when you dig through the MPC load screen, you'll see they're indeed gone. To get this preset data back, you will have to backup all your own data and then do a factory reset on your machine before moving the backed-up data back into the 5k (again, on your PC).
