You can use the samples and everything you have already picked out, you just need to get your mind into the "theme" first, than any sample you pick will have to contribute to the theme in you head. If you find the samples you have don't fit the theme then you may need to do some major distortion or flipping, but if that gets too involved just look at where you feel the "theme" is going...then work with that.
If you wanted to make a "johnny Mnemonic" themed tape, you can pick specific sub themes from the movie. Like make a beat themed like the crazy cyber-dolphin or the feeling of the growing NAS (comparable to a lot of modern diseases).
I think it's less important that you embody your source inspiration and more important that you create something that reflects what YOU think/feel about the source (which can be anything).
