What MIDI and or Audio interface are you using? That would be more likely the thing to troubleshoot before the OS itself. (Vista handles MIDI just like XP and 7/8 and I've never had that issue as an OS related one.)
Although there is one thing that can happen, not sure if it applies here. I think it's a limit of 16 MIDI entries that Windows will allow and write to the registry. When you swap to different USB ports with MIDI enabled gear (those random "why did the driver just re-install itself situations), it writes a new entry.
After the 16 are full it won't write new ones unless you manually delete the dupes or ones you no longer use (old gear maybe).
Not sure if that has any bearing on your issue but it's the only thing I know off on an OS level that will stop your MIDI broadcast dead in it's tracks.
I gotta run, but I'll come back and post how and where the settings are. (Or maybe Google Windows+MIDI registry+limits and you should find something if you want to investigate that).
But also please post more about your Studio One and MPC MIDI settings just so the forum can visaualize the "puzzle" better.
