I think I understand what you mean after taking a second look at your question. Quantize can kill the human feel of your beat - use it with care. When you say you play with Quantize on and off. I'm assuming you quantize some tracks and not others? That can work, but may not be enough to affect the overall feel. You could also trying playing without quantize on at all and then go fix some of your mistakes in the editor.
If you have to play with Quantize on, but your beat still sounds like a machine produced it, you can try a couple of things. Play with your velocities. Louder and softer hits mixed up creatively can give a more human feel. Mix up your beat. Repeat hits in some bars while removing them in others.
