the most important upgrade would probably be the memory expansion if it is not already in your mpc to get more sample time.
then some cf-cards to store your projects and samples or even a harddrive to store much more if it is not already in your mpc.
if you want to transfer big amounts of data to the mpc like sample libraries in gb size then it would be helpful to use a external hdd enclosure for that transfer process, because the usb on the mpc is only usb 1.0 and that is very slow!

You favor sampling, good. Now do yourself a further favor and read the manual. However confusing it might be, follow the pictures and do what it tells you and have fun with it. Ok, FUN! Too many dudes get themselves a sampler and get so serious about it that their beats sound all stiff and tight-ass like metrosexual wall-street lurches. Its not gonna yell at you, alright, its not gonna tell her friends what short and stubby fingers you got so get that shit out your mind sun-son.
A preamp and speakers is whats really essential. I have a line/mic type preamp. I send all my samples through it and sample from it and use it for vocals. It has EQ too.

How is Ur MP equiped? Where are you planning on getting your samples from? What have you? What do you want to do?
Vinyl?

do you have the CD drive Built in?
a mic...
Mixer-wise
a mixer should be helpfull. A DJ mixer ging to Ur MPs inputs. A mixer with 10 or more channels for Ur outputs.
You might want to learn about mixers.
The possibilities are endless.
