How many bars is your loop?
Did you trigger you loop at 001.01.01 so there is no pause in playing?
Maybe it will be easier to re-trigger your loop when you want it to play again ie, 2 bar loop, loop off, trigger at 001.01.01 and again at 003.01.01.
Do you have a space/gap/air in the front of the sample that would be the cause of your delayed playback problem?

The loop was on 4bars and there was no gap and yes it was at 001.01.01 I'm using a MPCXL with Pro Tools ,Trion LE,Famton XL module i trim it up and went to the loop screen looped it up tight so when i go to record it on a Seq with no gaps and i go to Seq 2 to add something else thats when i hear a pause

Maybe you should:
time stretch
tune your sample up or down (with the sequencer playing so everytime the loop starts back again you will hear the change you just made)
