You can just load a sample of a 440hz sine wave or any other reference tone you need.

i use a "tuner" program, which is just a sine wav inst pgm (or any of those "example synths" that murai put up) to tune my samples real carefully. i do this every time i chop a sample to get em as close to the "right" tuning as possible so it's easier to overlay stuff in reason/using inst pgms

Some audio editors can synthesize sounds. I know SoundForge does that. I used that to tune my bass, before I found an online tuner. Maybe wavosaur (freeware) does the trick too...
