No, a S5000 without an effects card isn't a factor worth worrying about, nor is upgrading from the stock 64 voice card either, you're only making hiphop after all. I'm not willing to spend upwards of 300+ for the EB20 when I could put that money to other much more interesting equipment. Get a better interface for recording your music, that's a great idea. You don't need the USB card either as I'm sure no normal person is willing to downgrade their operating system, or run a separate version of Windows in a Virtual Machine just to interface with it. (Unless I'm unaware if people are getting the USB card to work with Windows 10?) A SCSI CF card reader is the best option and there are plenty of utilities to take full .wav file sample sets and auto assign them to .akp files.
If your goal is to sound as close to the SP-1200/SP-12 with hardware, then finding an Emax 1 (not the Emax ) would be the route to take. They are however not affordable to get but thankfully the learning curve is almost nothing as the instructions are written right on the face plate.
When you start complicating things by adding hardware for 'effects' and 'filters', it only adds to convuse things in my opinion. Most of that sound that people are chasing from that boombap era (in my opinion) has to do with the way it was all recorded direct to tape and the associated artifacts of that. Not how many different pieces of rack gear for reverb effects they had running. Go get yourself a cheap Alesis Quadraverb series and choose one of the generic hall verb or delay settings.
Yes a lot of 'newer' E-mu series gear such as the E64 series, ESI-32, etc are cool and can be incorporated in modern DAW setups as all (as far as I remember) effects values can be controlled in real time over midi CC, and from minimal interaction I've had with the 32 and the E6400 ULTRA, I remember the midi CC was responding quite fast but that is a separate topic and doesn't really seem suited towards your interest in boom bap.
The thing I would suggest taking into consideration is how many pieces of gear do you think you need to essentially produce a rather simplistic form of hiphop. And that isn't even an insult to the boom bap sound, I believe you would be much happier by simply using your 2000XL and getting an 404 and using the Lo-Fi setting on it and see where that gets you before moving into the realm of rack mount equipment.
